Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

The full-length cDNA ORF (1-391aa) of Enterobacteria phage T4 (Bacteriophage T4) Recombination and repair protein (UVSX) was N-terminally tagged a 6xHis-SUMO and then expressed in E.coli. The Recombinant Enterobacteria phage T4 UVSX protein thus obtained was purified using SDS-PAGE analysis and got over 90% of purity. Under reducing conditions, the UVSX protein migrated to an approximately 61 kDa molecular mass band on the SDS-PAGE gel. This protein was also validated by the LC-MS/MS Analysis. UVSX proteins in-stock are available now. This recombinant UVSX protein may be the immunogen for specific antibody production or in the studies of homologous recombination (HR).

UVSX is the recombinase of the HR machinery. HR plays an important role in repairing the error-free of DNA double-strand breaks and restarting the stagnant replication fork. UVSX is important for the late replication stage of the T4 phage, during which replication initiates by recombination. Together with other recombinases, UVSX forms presynaptic filaments on ssDNA (single-stranded DNA) that are activated to search for homology in dsDNA (double-stranded DNA) and to exchange DNA strand.

 Q&A
Q:

Hello, can I know the concentration of this protein? Thanks.

A:
Thanks for your inquiry. We have 2 lots in stock, one lot is 0.2mg/ml, the other lot is 0.4mg/ml. If you have special request on the concentration , please let us know.
Q:

Today I received 2 proteins with same catalog# CSB-EP361289EDZ but one of them was ordered with Endotoxin removal and the other one was ordered with no extra services, they both are the same host, lot# , exp date ,conc and vol on the label.
I have the two different data sheets but how am I going to able to distinguish the proteins? Please let me know what I can do.

A:
Thanks for your feedback.1. The naming of the documents are different, one is "CSB-EP361289EDZ-Lot03419 Datasheet&COA", and the other is "CSB-EP361289EDZ-Lot03419 Datasheet&COA -Endotoxin Removal".
2. If we have endotoxin removal, we will remark "<1.0 EU per 1μg of the protein by the LAL method. pass" in following field in COA.
If we haven't performed endotoxin removal, we will remark "Untreated" in following field in COA.

Target Background

Function
Important in genetic recombination, DNA repair, and replication. Possesses pairing and strand-transfer activity. Interacts with dda and gene 32 proteins.
Gene References into Functions
  1. the uvsX mutant executes recombination-dependent replication but does it by a qualitatively different way. PMID: 22365497
  2. The authors show that the phage helicase UvsW completes the UvsX-promoted strand-exchange reaction, allowing the generation of a simple nicked circular product rather than complex networks of partially exchanged substrates. PMID: 21035462
  3. UvsX suffice to rescue stalled T4 replication forks in vitro PMID: 15194689
  4. allosteric switch residue His195 controls the formation of an asymmetric conformation of UvsX-ssDNA filaments that is active in DNA strand exchange. PMID: 19027026
  5. unlike wild-type UvsX, the ssDNA-dependent ATPase activities of UvsX-H195Q/A are strongly inhibited by both low and high concentrations of UvsY protein. PMID: 19244311
