Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

 Q&A
Q:

Because this protein is a several-times transmembrane protein, is there any particular reason why it isn't being expressed by the cell-free system? I was wondering if the protein should be provided in non-ionic detergent such as 0.03% n-OPE. What are your thoughts?

A:
Very nice to receive your inquiry.
Yes, you're right, this is several-times transmembrane protein, but this is a self protein of E.coli, Literature for your reference: https://amb-express.springeropen.com/articles/10.1186/s13568-017-0452-8
In addition, we have already successfully developed some related proteins, please check them as follows, so this is just a special case that we choose the regular four expression systems to express this several-times transmembrane protein, becasue we think there is no difficulty to express this protein especially with E.coli expression system.
In addition, we evaluate that if choosing cell free expression system, the cost is relatively high and the yield will be very low, so we don't think it's a good choice.
The general detergent of transmembrane proteins will be depended on the specific circumstances, but we generally use anionic surfactants for E.coli system, such as: skl (alias: sodium lauroylsarcosine; Sodium N-lauroylsarcosine;), or Non-ionic surfactants such as tritonx-100. However, you are assured that the final product form we provide is soluble.

Q:

Could you please let me know the exact composition of the Tris-based buffer for this product? Thanks for your help.

A:
Thanks for your inquiry. The composition of the Tris-based buffer is 10 mM Tris-HCl, 1 mM EDTA, pH 8.0, 50% glycerol. Have a nice day~

Target Background

Function
Forms pores that allow passive diffusion of small molecules across the outer membrane.; (Microbial infection) Supports colicin E5 entry in the absence of its major receptor OmpF.; (Microbial infection) A mixed OmpC-OmpF heterotrimer is the outer membrane receptor for toxin CdiA-EC536; polymorphisms in extracellular loops 4 and 5 of OmpC confer susceptibility to CdiA-EC536-mediated toxicity.
Gene References into Functions
  1. Studied the rate of entry (association) and exit (dissociation) of six antibiotics to/from reconstituted E. coli porin OmpC. WaterMap was used to estimate the total water transfer energies resulting from transient occupation by each antibiotic. PMID: 23011778
  2. X ray structure of OmpC complexed with lactoferrin PMID: 16511154
Subcellular Location
Cell outer membrane; Multi-pass membrane protein.
