Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

Recombinant Helicobacter pylori Vacuolating cytotoxin autotransporter(vacA) is a partial-length protein derived from E.coli. The expression region of this protein is the 1018-1290aa of Helicobacter pylori vacA. It carries an N-terminal 6xHis-B2M-tag and has a calculated molecular weight of 43.6 kDa. Its purity is greater than 90% as determined by SDS-PAGE and its identity was validated by the LC-MS/MS Analysis. This recombinant vacA protein may be used to generate specific antibodies and in microbiology research.

VacA is one of the predominant protein toxins secreted by the type V auto-transport secretion system of the Helicobacter pylori (H. pylori). The H. pylori is a pathogenic bacterium that potentially causes peptic ulcers of the human stomach even gastric cancer. VacA is important for H. pylori colonization in vivo and has the capacity of causing "vacuole"-like membrane vesicles in the cytoplasm of gastric cells. Besides, it has other effects on target cells, including disturbing mitochondrial functions, stimulating apoptosis, and blocking T cell proliferation.

Target Background

Function
Induces vacuolation of eukaryotic cells. Causes ulceration and gastric lesions.
Gene References into Functions
  1. Helicobacter pylori VacA induces autophagic cell death in gastric epithelial cells via the endoplasmic reticulum stress pathway. PMID: 29238039
  2. The results indicate that cortactin is involved in the regulation of apoptosis induced by VacA in gastric cells. PMID: 26289258
  3. Genotype testing of vacA s- and m- region will be useful in screening susceptible individuals for duodenal ulcer development. PMID: 25063579
  4. These results suggested that cagA and vacA gene expression is upregulated in Helicobacter pylori, especially by host cell contact, and Fur has a role in the upregulation. PMID: 24020886
Subcellular Location
[Vacuolating cytotoxin autotransporter]: Periplasm.; [Vacuolating cytotoxin]: Secreted. Cell surface.; [Vacuolating cytotoxin translocator]: Cell outer membrane; Multi-pass membrane protein.
