Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

The recombinant Human AP1B1 was expressed with the amino acid range of 1-584. This AP1B1 protein is expected to have a theoretical molecular weight of 73.3 kDa. This AP1B1 recombinant protein is manufactured in e.coli. Fusion of the N-terminal 6xHis tag and C-terminal Myc tag into the AP1B1 encoding gene fragment was conducted, allowing for easier detection and purification of the AP1B1 protein in subsequent stages.

The human AP-1 complex subunit beta-1 (AP1B1) is a crucial component of the adaptor protein complex 1 (AP-1), which plays a fundamental role in intracellular protein trafficking. AP1B1 is specifically associated with the basolateral sorting of membrane proteins in polarized cells. It aids in cargo selection, vesicle formation, and transport between the trans-Golgi network and endosomes. In cell biology and membrane trafficking research, AP1B1 is studied for its involvement in maintaining cellular organization and polarity. Understanding its functions contributes to insights into intracellular transport mechanisms and potential implications for various physiological processes.

Target Background

Function
Subunit of clathrin-associated adaptor protein complex 1 that plays a role in protein sorting in the late-Golgi/trans-Golgi network (TGN) and/or endosomes. The AP complexes mediate both the recruitment of clathrin to membranes and the recognition of sorting signals within the cytosolic tails of transmembrane cargo molecules.
Gene References into Functions
  1. both ubiquitous (AP-1A) and epithelium-specific (AP-1B) forms of the tetrameric clathrin adaptor AP-1 are capable of carrying out basolateral sorting of ClC-2. PMID: 25739457
  2. AP1B regulates basolateral EGFR membrane delivery predominantly in the biosynthetic route in MDCK cells monolayers. PMID: 23205726
  3. The conclude that in addition to its role in endocytosis, ARH cooperates with AP-1B in basolateral exocytosis of LDLR from recycling endosomes. PMID: 21444685
Subcellular Location
Golgi apparatus. Cytoplasmic vesicle, clathrin-coated vesicle membrane; Peripheral membrane protein; Cytoplasmic side. Note=Component of the coat surrounding the cytoplasmic face of coated vesicles located at the Golgi complex.
Protein Families
Adaptor complexes large subunit family
Tissue Specificity
Widely expressed.
