There are some publications that show that E. coli have arginine deiminases. However, I am also using a protein produced by a collaborator that also used E. coli but with a transformation buffer with 20 mM CaCl2. This protein shows a high amount of basal citrullination. Do you use an expression protocol that avoids activation of arginine deiminases (e.g., with low calcium)?
We didn't use an expression protocol that avoids activation of arginine deiminases, and we didn't add any calcium.
This protein contains a His-SUMO tag, correct? We are using it in an assay where the His tag is used to immobilize the protein. The binding seems to be not as strong comparatively or it takes a longer time.
Yes, the tag type for CSB-EP014597HU(A4) is N-terminal 6xHis-SUMO-tagged.
This protein is purified by Ni column affinity purification. We used his tag for binding, there should be no problem with binding.
