The generation of the recombinant human cation-independent mannose-6-phosphate receptor (IGF2R) with a C-terminal 6xHis-Myc-tag is accomplished using an efficient yeast expression system. The process involves cloning the gene fragment encoding the 628-772aa of the human IGF2R into a specialized expression vector. This vector contains a C-terminal 6xHis-Myc-tag, which is designed to facilitate easy purification and detection of the recombinant protein. After introducing the constructed vector into yeast cells through transfection, transfected cells are selected. Large-scale culture of the transfected yeast cells is established to allow for optimal protein expression. The expressed recombinant IGF2R protein is then harvested from the cell lysate and subjected to purification techniques, ensuring a high level of purity. SDS-PAGE analysis confirms the protein's purity, which exceeds 90%.
