Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Description

Dipeptidyl peptidase 1 functions as a key activator of granule-associated serine proteases in immune and inflammatory cells, making its catalytic properties central to understanding neutrophil and cytotoxic lymphocyte biology. This full-length mature protein (aa 25–463) demonstrates specific activity exceeding 200 pmol/min/μg against the fluorogenic substrate Gly-Arg-AMC following activation by cathepsin L, providing a quantitative basis for kinetic parameter analysis including Km and Vmax determination, substrate specificity profiling, and inhibitor screening with IC50 measurements. The C-terminal His tag preserves access to the active site while enabling straightforward purification, and purity greater than 95% combined with endotoxin levels below 1.0 EU/μg meets the quality thresholds commonly required for accurate enzymatic assays and drug candidate evaluation studies where contaminating proteases or endotoxin could confound inhibition mechanism analysis.

Thiol protease. Has dipeptidylpeptidase activity. Active against a broad range of dipeptide substrates composed of both polar and hydrophobic amino acids. Proline cannot occupy the P1 position and arginine cannot occupy the P2 position of the substrate. Can act as both an exopeptidase and endopeptidase. Activates serine proteases such as elastase, cathepsin G and granzymes A and B. Can also activate neuraminidase and factor XIII.
