Description |
The production of this recombinant Human GM2A protein is just like all recombinant proteins. The process involved transfecting E.coli cells with DNA vector containing the template of recombinant DNA. The E.coli cells containing the template were then cultured so that they could transcribe and translate the GM2A protein. N-terminal 6xHis-SUMO tag was used in the process. The purity is 85% determined by SDS-PAGE. (The activity of this GM2A protein has been validated.) The GM2A gene provides instructions for making a protein named ganglioside GM2 activator (also called cerebroside sulfate activator protein (GM2AP), sphingolipid activator protein 3 (SAP3)). The GM2A protein is involved in various biological processes, including ganglioside catabolic process, lipid storage and transport, neuromuscular process controlling balance, etc. The mature form of ganglioside GM2 activator protein is a small (20 kDa), a heat-stable, long-lived, protease-resistant glycosylated protein that normally resides in the lysozyme. |

Reconstitution | We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference. |

Function |
The large binding pocket can accommodate several single chain phospholipids and fatty acids, GM2A also exhibits some calcium-independent phospholipase activity. Binds gangliosides and stimulates ganglioside GM2 degradation. It stimulates only the breakdown of ganglioside GM2 and glycolipid GA2 by beta-hexosaminidase A. It extracts single GM2 molecules from membranes and presents them in soluble form to beta-hexosaminidase A for cleavage of N-acetyl-D-galactosamine and conversion to GM3. Has cholesterol transfer activity.
