Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Description

IL-1 receptor type 2 functions as a decoy receptor that sequesters IL-1β without triggering downstream signaling, making it a critical negative regulator in inflammatory cascades and a target for therapeutic modulation in autoimmune and inflammatory diseases. This mammalian-expressed construct spanning residues 14–343 captures the complete extracellular ligand-binding domain, ensuring native glycosylation patterns that preserve conformational epitopes essential for accurate receptor-ligand interaction studies. Functional ELISA validation demonstrates specific binding to immobilized IL-1β with an EC50 of 89.65–100.5 ng/mL, providing quantitative evidence that this protein supports ligand-binding assays, competitive inhibition screens for IL-1 pathway modulators, and blocking antibody characterization in drug discovery programs targeting inflammatory signaling. The combination of >95% purity by SDS-PAGE, >90% purity by SEC-HPLC, and endotoxin levels below 1.0 EU/μg satisfies the quality criteria typical for surface plasmon resonance affinity measurements, bio-layer interferometry kinetic studies, and therapeutic antibody epitope mapping workflows.

Function

Non-signaling receptor for IL1A, IL1B and IL1RN. Reduces IL1B activities. Serves as a decoy receptor by competetive binding to IL1B and preventing its binding to IL1R1. Also modulates cellular response through non-signaling association with IL1RAP after binding to IL1B. IL1R2 (membrane and secreted forms) preferentially binds IL1B and poorly IL1A and IL1RN. The secreted IL1R2 recruits secreted IL1RAP with high affinity; this complex formation may be the dominant mechanism for neutralization of IL1B by secreted/soluble receptors.
