Description

The plasmid vector incorporates the gene encoding the Human SMAD6 protein (1-496aa), generating recombinant plasmid, which is then introduced into baculovirus cells. Selection of positive baculovirus cells is based on their capacity to withstand a particular antibiotic. Subsequently, the baculovirus cells containing the recombinant plasmid are cultivated under conditions that facilitate the expression of the Human SMAD6 gene. The protein carries a N-terminal 10xHis tag and C-terminal Myc tag. Following expression, affinity purification is employed to isolate and purify the recombinant Human SMAD6 protein from the cell lysate. Denaturing SDS-PAGE is used to resolve the resulting recombinant protein, enabling an estimation of its purity, exceeding 85%.

Transforming growth factor-beta superfamily receptors signaling occurs through the Smad family of intracellular mediators. SMAD6 is an inhibitory Smad (i-Smad) that negatively regulates signaling downstream of type I transforming growth factor-beta. Acts as a mediator of TGF-beta and BMP anti-inflammatory activities. Suppresses IL1R-TLR signaling through its direct interaction with PEL1, preventing NF-kappa-B activation, nuclear transport and NF-kappa-B-mediated expression of proinflammatory genes. Blocks the BMP-SMAD1 signaling pathway by competing with SMAD4 for receptor-activated SMAD1-binding. Binds to regulatory elements in target promoter regions.
