SMAD6

SMAD6 (SMAD family member 6), also known as MADH6 or JV18-1, is an inhibitory SMAD protein. It negatively regulates TGF-β superfamily signaling—especially BMP pathways—by competing with receptor-regulated SMADs (SMAD1/5/8) for type I BMP receptor binding or sequestering SMAD4 to block nuclear translocation of active signaling complexes.
Dysregulation of SMAD6 links to skeletal disorders (e.g., craniosynostosis), cardiovascular diseases (aortic aneurysms), and some cancers (e.g., colorectal cancer, where it may act as a tumor suppressor). Current drug development focuses on modulating BMP/TGF-β pathways; direct SMAD6-targeted agents remain in preclinical stages, aiming to correct signaling imbalances in these conditions.
