Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Description

Netrin-1 orchestrates axon guidance, angiogenesis, and tumor cell migration through interactions with DCC and UNC5 family receptors, making it a pivotal target in neurodevelopmental and cancer biology research. This mammalian-expressed protein spanning residues 22–604 captures the full secreted form and demonstrates robust receptor-binding activity, with an EC50 of 26.43–30.77 ng/mL in functional ELISA against anti-NTN1 antibody, providing a quantitative benchmark for receptor-ligand binding and competition assays as well as antibody characterization studies. The mammalian expression system preserves native glycosylation patterns critical for proper folding and receptor engagement, supporting use in cell migration assays, angiogenesis models, and tumor invasion studies where post-translational modifications influence biological activity. Purity exceeding 95% by SDS-PAGE and endotoxin levels below 1.0 EU/μg satisfy the quality thresholds commonly required for sensitive cell-based assays and in vivo tumor biology models.

Function

Netrins control guidance of CNS commissural axons and peripheral motor axons. Its association with either DCC or some UNC5 receptors will lead to axon attraction or repulsion, respectively. Binding to UNC5C might cause dissociation of UNC5C from polymerized TUBB3 in microtubules and thereby lead to increased microtubule dynamics and axon repulsion. Involved in dorsal root ganglion axon projection towards the spinal cord. It also serves as a survival factor via its association with its receptors which prevent the initiation of apoptosis. Involved in tumorigenesis by regulating apoptosis.
