Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
13-23 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

The production of the recombinant human O-phosphoseryl-tRNA(Sec) selenium transferase (SEPSECS) follows a well-defined process within the E. coli expression system. Initially, the gene fragment encoding the full-length SEPSECS protein (amino acids 1-501) is inserted into a suitable expression vector. This vector is designed to include an N-terminal 10xHis-tag and a C-terminal Myc-tag, which aids in the purification and detection of the recombinant protein. Through transformation, the recombinant expression vector is introduced into E. coli cells, followed by the selection of the transformed cells. Subsequently, the transformed cells are cultured in an optimized growth medium to facilitate efficient protein expression. Induction of protein production is achieved by the addition of a specific inducer, such as IPTG. The cells are harvested and lysed to release the intracellular contents containing the recombinant SEPSECS protein. The purity of the recombinant SEPSECS protein is rigorously assessed through SDS-PAGE, reaching up to 90%.

Target Background

Function
Converts O-phosphoseryl-tRNA(Sec) to selenocysteinyl-tRNA(Sec) required for selenoprotein biosynthesis.
Gene References into Functions
  1. Structural basis for early-onset neurological disorders caused by mutations in human selenocysteine synthase has been described. PMID: 27576344
  2. Epileptic encephalopathy with burst suppression without brain malformations is associated with pathogenic variation in SEPSECS. PMID: 28133863
  3. The progression of clinical symptoms in these families is evidently slower than in previously reported cases, and the cerebellar atrophy milder by brain MRI, indicating that SEPSECS mutations are also involved in milder late-onset cerebellar atrophy. PMID: 26888482
  4. results suggest SEPSECS as a candidate gene for progressive encephalopathies with lactate elevation. PMID: 26115735
  5. structural analysis of the terminal catalytic complex in selenocysteine synthesis PMID: 25190812
  6. Data suggest SEPSECS silencing in placental trophoblasts inhibits proliferation, induces apoptosis, and reduces production of progesterone/chorionic gonadotropin (P/hCG); SEPSECS over-expression promotes cell proliferation and secretion of P/hCG. PMID: 23966103
  7. The human SepSecS protein is also known as soluble liver antigen/liver pancreas (SLA/LP), which represents one of the antigens of autoimmune hepatitis. PMID: 20623998
  8. SepSecS mutations cause autosomal-recessive progressive cerebellocerebral atrophy in Jews of Iraqi and Moroccan ancestry. PMID: 20920667
  9. detectable in liver extracts of autoimmune hepatitis patients' cells as the major autoantigenic component PMID: 19683415
  10. crystal structure of tRNA(Sec) in complex with SepSecS, phosphoserine, and thiophosphate, together with in vivo and in vitro enzyme assays, supports a pyridoxal phosphate-dependent mechanism of Sec-tRNA(Sec) formation PMID: 19608919
