Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Description

The construction of a plasmid coding for the Lassa virus (strain GA391) GPC protein (59-258aa) is the initial step for the preparation of the recombiant Lassa virus (strain GA391) GPC protein. The next is to transform the constructed plasmid into e.coli cells. e.coli cells containing the plasmid are screened and then cultured under conditions that promote the expression of the gene of interest. The protein is equipped with a N-terminal 10xHis tag and C-terminal Myc tag. After that, affinity purification is used to isolate and purify the recombinant GPC protein from the cell lysate. Finally, the resulting recombinant GPC protein undergoes SDS-PAGE analysis, demonstrating a purity greater than 90%.

Function

class I viral fusion protein that directs fusion of viral and host endosomal membranes, leading to delivery of the nucleocapsid into the cytoplasm. Membrane fusion is mediated by irreversible conformational changes induced upon acidification in the endosome.; Stable signal peptide (SSP): cleaved and functions as a signal peptide. In addition, it is also retained as the third component of the GP complex. The SSP is required for efficient glycoprotein expression, post-translational maturation cleavage of GP1 and GP2, glycoprotein transport to the cell surface plasma membrane, formation of infectious virus particles, and acid pH-dependent glycoprotein-mediated cell fusion.; interacts with the host receptor.
