SIGLEC9 mediates immune cell inhibitory signaling through recognition of sialylated glycans, making it a target of interest in immunomodulation and inflammation research. This cynomolgus macaque ortholog, spanning residues 20–350 and expressed in mammalian cells to preserve native glycosylation and folding, demonstrates robust binding to an anti-SIGLEC9 recombinant antibody with an EC50 of 7.698–8.925 ng/mL in functional ELISA, confirming conformational integrity of the extracellular domain. Surface plasmon resonance analysis further validates high-affinity interaction, yielding a KD of 4.34 nM, which supports use in therapeutic antibody epitope mapping, competitive inhibition screening, and affinity characterization by SPR or biolayer interferometry.
