Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Small-conductance calcium-activated nonselective cation (SCAN) channel which acts as a regulator of phospholipid scrambling in platelets, osteoblasts and fetal thymocytes. Phospholipid scrambling results in surface exposure of phosphatidylserine which in platelets is essential to trigger the clotting system whereas in osteoblasts is essential for the deposition of hydroxyapatite during bone mineralization. Has calcium-dependent phospholipid scramblase activity; scrambles phosphatidylserine, phosphatidylcholine and galactosylceramide. Can generate outwardly rectifying chloride channel currents in airway epithelial cells and Jurkat T lymphocytes.
Gene References into Functions
  1. TMEM16F knockout did not affect Ca2+ signaling and Ca2+-activated whole cell currents in podocytes. TMEM16F shows little effect on basal properties of podocytes and does not appear to be essential for renal function. PMID: 29912162
  2. TMEM16F interaction with phosphatidylinositol-(4, 5)-bisphosphate works synergistically with membrane depolarization to facilitate Ca(2+)-gating of TMEM16F. PMID: 29382763
  3. TMEM16F is an essential component of the microglial response to injury and suggest the importance of microglial phagocytosis in the pathogenesis of neuropathic pain. PMID: 27332874
  4. The present data demonstrate the activation of ANO6 during necroptosis, which, however, is not essential for cell death. PMID: 27535660
  5. TMEM16F deficiency results in sustained signaling and augmented T cell activation. PMID: 27810927
  6. Platelet TMEM16F activity contributes significantly to hemostasis and thrombosis but not cerebral thromboinflammation. PMID: 27634832
  7. ANO6 produces volume-regulated anion currents and controls cell volume in four unrelated cell types. PMID: 26438191
  8. deficiency in Ano6 resulted in reduced viability with increased bleeding time PMID: 26481309
  9. Although broadly expressed, Ano6 has no role in intestinal cholinergic Cl- secretion. PMID: 24974903
  10. Ano6 mediates effects essential for innate immunity downstream of P2X7 receptors in macrophages. PMID: 25651887
  11. TMEM16F is required for phosphatidylserine exposure and microparticle release in activated mouse platelets. PMID: 26417084
  12. Ano6 and Ano2 proteins co-localize in the ciliary membrane microdomains, which indicates a role for Ano6 in olfactory signaling. PMID: 25500808
  13. human osteoblasts and osteoblasts from Ano6(-/-) and WT mice, we demonstrate that NCX1 requires Ano6 to efficiently translocate Ca(2+) out of osteoblasts into the calcifying bone matrix PMID: 25589784
  14. Ano6 plays an essential role in C2C12 myoblast proliferation PMID: 24663380
  15. The mTMEM16F-associated ICl,Ca was abolished in one mutant of the putative pore region (R592E. PMID: 23630341
  16. In primary Ano6(-/-) osteoblasts, mineralization is delayed, indicating a cell autonomous function of Ano6. PMID: 22936354
  17. TMEM16F forms a SCAN channel with considerable Ca2+ selectivity-an unprecedented finding of a cation channel as a closely related family member of anion channels; loss of TMEM16F function greatly reduced the propensity to thrombosis. PMID: 23021219
  18. Data show the expression of Tmem16f during murine embryogenesis with an emphasis on the respiratory, digestive, skeletal, and integumentary systems. PMID: 18729231
  19. during mouse cephalic development: detected in subventricular zone and differentiating fields, in rostral spinal cord motor neuron area, stria vascularis,in submandibular autonomic ganglia, in palate, tooth and tongue, and in osteoblasts and chondrocytes PMID: 19059364
