ANO6

Anoctamin 6 (ANO6), also known as transmembrane protein 16F (TMEM16F), is a calcium-activated chloride channel and phospholipid scramblase. It mediates bidirectional phospholipid translocation across the plasma membrane, particularly exposing phosphatidylserine (PS) on the cell surface under calcium stimulation— a process critical for blood coagulation, apoptotic cell clearance, and cell signaling.
Dysfunction of ANO6 is associated with Scott syndrome, a rare bleeding disorder marked by impaired platelet PS exposure. Overexpression of ANO6 is linked to certain cancers, promoting cell migration and metastasis. Preclinical research is exploring small-molecule inhibitors targeting its channel or scramblase activity as potential therapies for thrombotic diseases or ANO6-driven malignancies, though clinical trials are still in early stages.
