Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Description

Glucagon receptor signaling drives hepatic glucose output and represents a validated target in metabolic disease and certain cancers where aberrant glucagon pathways fuel tumor growth. This mammalian-expressed construct spanning residues 27–137 captures the extracellular ligand-binding domain with native post-translational modifications that preserve conformational epitopes, as demonstrated by its ability to bind an anti-Gcgr recombinant antibody with an EC50 of 29.63–35.34 ng/mL in functional ELISA. The quantified binding kinetics support use in competitive inhibition assays, therapeutic antibody epitope mapping, and small-molecule or biologic inhibitor screening campaigns where accurate receptor-ligand interaction data are essential. Purity exceeding 90% by SDS-PAGE and endotoxin below 1.0 EU/μg align with standards expected in SPR, BLI, and cell-based assays requiring low background interference.

Function

G-protein coupled receptor for glucagon that plays a central role in the regulation of blood glucose levels and glucose homeostasis. Regulates the rate of hepatic glucose production by promoting glycogen hydrolysis and gluconeogenesis. Plays an important role in mediating the responses to fasting. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of down-stream effectors, such as adenylate cyclase. Promotes activation of adenylate cyclase. Besides, plays a role in signaling via a phosphatidylinositol-calcium second messenger system.
