Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Key regulator of protein phosphatase 1C (PPP1C). Mediates binding to myosin. As part of the PPP1C complex, involved in dephosphorylation of PLK1. Capable of inhibiting HIF1AN-dependent suppression of HIF1A activity.
Gene References into Functions
  1. Phos-tag SDS-PAGE thus provides a suitable and convenient method for analysis of the complex, multisite MYPT1 phosphorylation events involved in the regulation of myosin light chain phosphatase activity and smooth muscle contraction. PMID: 26864694
  2. Both atorvastatin (10(-4) m) and Y-27632 (10(-6) m) pretreatment inhibited the phosphorylation of myosin phosphatase target subunit-1 (MYPT-1). PMID: 26610064
  3. EGF induces Ca(2+) sensitization in vascular smooth muscle by Rho-kinase-dependent inactivation of MLCP mediated by the EGF receptor/MEK/Erk1/2 pathway PMID: 26004531
  4. nuclear translocation of CaMKIIdelta3 following dephosphorylation at Ser(332) by PP1 likely accounts for BDNF expression and subsequent neurite extension and survival of dopaminergic neurons. PMID: 26163515
  5. These results indicate that PPP1R12A indeed plays a role in skeletal muscle insulin signaling PMID: 24972320
  6. During acute myocardial ischemia MYPT1 is targeted for degradation or proteolysis. Reduced MYPT1 expression results in a decrease in both tone and the sensitivity to NO-mediated SM relaxation of the peripheral vasculature. PMID: 23172397
  7. In healthy young female rats, MLCK activity is higher and MLCP activity is lower in the bladder relative to the urethra PMID: 21070574
  8. after hypoxia, increased myosin phosphatase target subunit 1 and myofibrillar protein phosphatase-1 activity impair aortic vasoreactivity through enhanced dephosphorylation of myosin light chain 20-kDa PMID: 12595284
  9. Alteration of MYPT1 expression was part of a complex change in protein expression that can be generalized as a modulation from a phasic (fast) to a tonic (slow) contractile phenotype. PMID: 14704233
  10. Congestive heart failure is associated with a decrease in the relative expression of the LZ+ MYPT1 isoform. Resistance to NO-mediated relaxation seen during CHF is a consequence of the decrease in the expression of the LZ+ MYPT1 isoform in smooth muscle. PMID: 15321927
  11. pathway for the dynamic modulation of myosin light chain 2 phosphorylation through receptor-dependent phosphorylation of MYPT PMID: 15908465
  12. C-terminus functions as an auto-inhibitory domain, and a central domain can mediate extensive reorganization of the actin cytoskeleton. PMID: 16106448
  13. Phosphorylation of Thr695 and Thr850 on MYPT1 was detected in A7r5 cells, but Thr850 was preferred by Rho-kinase and Thr695 was phosphorylated by an unidentified kinase(s). PMID: 16297917
  14. angiotensin converting enzyme inhibition prevents MYPT1 isoform switching, which would preserve normal flow, or NO-mediated vasodilatation in rats PMID: 16815432
  15. There is a rapid reduction in MYPT1 expression in the small resistance mesenteric artery in a disease model of altered blood flow. PMID: 17293476
  16. These results indicate that heat shock augments MYPT1 phosphorylation resulting in augmented vascular contraction. PMID: 17382904
  17. Treatment with flavone inhibits vascular smooth muscle contraction by decreasing the phosphorylation of the MYPT1. PMID: 17880363
  18. A new regulatory mechanism of myosin light-chain phosphatase via the interaction between 14-3-3 and MYPT1, is reported. PMID: 18094049
  19. We propose that MYPT1 isoform switching is an adaptive response to reduce vascular resistance and maintain uterine blood flow in the setting of hypertension-triggered inward remodeling of the UAs in hypertension of pregnancy. PMID: 18094148
  20. Ca2+ sensitization via phosphorylation of MYPT1 threonine-855 by Rho kinase contributes to the arterial myogenic response. PMID: 19359365
