GPA33 has high homology with CD2 and possesses a V-type Ig-like domain at its N terminus. It's also linked to T cell activation inhibitory proteins including CD276 (B7-H3) and VSIG4, as well as many cell adhesion molecules like CEACAM, ICAM, and NCAM (3D-Protein BLAST, NCBI). Immune dysfunction has been linked to GPA33. Polymorphisms in the gene encoding GPA33 have been linked to the autoimmune condition eosinophilic granulomatosis with polyangiitis. Furthermore, the GPA33 gene is found in a Sjögren's Syndrome susceptibility locus. Hypersensitivity to dietary allergens and increased inflammatory responses to experimentally caused colitis was seen in a GPA33 knockout mice model.
The preparation of the recombinant GPA33 antibody involves the mammalian cell lines expression of plasma vectors containing GPA33 antibody genes. B cells isolated from immunized animals' blood were treated to obtain RNA, which underwent reverse transcription to yield DNA genes. Antibody genes were sequenced and screened from the DNA. After transient expression, cell supernatant was collected and then purified using Affinity-chromatography to obtain the recombinant GPA33 antibody.
