  • IHC image of CSB-RA245537A0HU diluted at 1:100 and staining in paraffin-embedded human small intestine tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.

Description

GPA33, also known as glycoprotein A33, is a transmembrane protein predominantly expressed in normal intestinal epithelium and maintained in over 95% of colorectal carcinomas. This restricted expression pattern, combined with its stable presence on the cell surface of malignant cells, has established GPA33 as a compelling target for colorectal cancer research and therapeutic antibody development. Understanding GPA33 expression and localization provides valuable insights into intestinal biology and colorectal cancer progression.

This recombinant monoclonal antibody, clone 4H4, offers the reproducibility and consistency that demanding research applications require. Generated against a synthetic peptide derived from human GPA33 and produced using recombinant technology, this antibody delivers sequence-defined specificity with minimal lot-to-lot variation. For researchers conducting longitudinal studies or multi-site collaborations, this consistency translates directly into comparable results across experiments and reduced time spent on antibody re-optimization.

Validation studies demonstrate reliable performance in immunohistochemistry applications on human tissues. Testing on paraffin-embedded human small intestine tissue confirms detection of GPA33 in its expected physiological context, while staining of human colon cancer tissue validates utility in oncology research settings. Both applications performed optimally at 1:100 dilution using citrate buffer antigen retrieval, providing a solid starting point for your experimental optimization.

The antibody is supplied in a glycerol-containing buffer formulation designed for long-term stability at -20°C or -80°C, supporting extended research programs. Whether you are investigating intestinal epithelial biology, characterizing colorectal tumor samples, or evaluating GPA33 as a therapeutic target, this recombinant antibody provides the specificity and consistency essential for meaningful immunological and cancer research outcomes.

  2. A33 expression is directly correlated to colorectal tumor differentiation. PMID: 27272411
  3. Data indicate that EpCAM- and A33-Exos were released from the apical and basolateral surfaces, respectively. PMID: 23230278
  4. Data indicate positive staining was observed for both A33 expression and KRN330 binding from patients biopsied on Day 8, 9 or 14 following initial dosing of 3 mg/kg. PMID: 23294608
  5. The A33-dependent incorporation of B5 into extracellular enveloped vaccinia virions is mediated through an interaction between their lumenal domains. PMID: 22623777
  6. Increased interaction between vaccinia virus proteins A33 and B5 is detrimental to infectious extracellular enveloped virion production. PMID: 22623782
  7. cloning, chromosomal localizations, exon-intron structures and transcription start sites - target gene for the intestine-specific homeobox transcription factor, CDX1. PMID: 12114523
