  • IHC image of CSB-RA179147A0HU diluted at 1:100 and staining in paraffin-embedded human melanoma cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.
  • Overlay Peak curve showing HepG2 cells stained with CSB-RA179147A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10, 000 events was performed.

Description

HAPLN1, also known as cartilage link protein 1, plays a fundamental role in extracellular matrix organization by stabilizing the interaction between hyaluronan and proteoglycans such as aggrecan and versican. This stabilization is essential for maintaining the structural integrity of cartilage and other connective tissues, making HAPLN1 a protein of significant interest in research areas spanning cartilage biology, tissue development, and signal transduction pathways that govern matrix remodeling.

This recombinant rabbit monoclonal antibody, clone 8F4, offers the consistency and reproducibility that demanding experimental workflows require. Because it is generated from a defined sequence rather than traditional hybridoma methods, researchers benefit from lot-to-lot uniformity, ensuring that results remain comparable across extended studies and collaborative projects. The antibody was raised against a synthetic peptide derived from human HAPLN1 and demonstrates cross-species reactivity with human, mouse, and rat samples, providing flexibility for comparative studies and translational research models.

Validation across multiple platforms confirms the antibody's versatility. Western blot analysis detects HAPLN1 in diverse human cell lines including COLO205, MCF7, U87, SH-SY5Y, and A2780, as well as in mouse and rat brain tissue lysates. The observed band at approximately 48 kDa, slightly higher than the predicted 40 kDa molecular weight, likely reflects post-translational modifications such as glycosylation, which is characteristic of this secreted extracellular matrix protein. Immunohistochemistry validation in human melanoma tissue and flow cytometry analysis in HepG2 cells further demonstrate reliable detection in both fixed tissue and cell-based applications.

Whether investigating matrix biology, cartilage pathology, or signal transduction mechanisms, this antibody provides a dependable tool for characterizing HAPLN1 expression and localization.
