We established a cell type-resolved prognostic atlas for PDAC using snRNA-seq of 152 patients, identifying thousands of cell-type-specific prognostic genes and bidirectional effects. Spatial transcriptomics revealed a therapy-resistant niche. PLOD2 was validated as a pan-cell-type poor-prognosis target, and PROTAC-G3 effectively inhibited tumors. Bidirectional Ephb4-ephrinb2 signaling induces anti-ferroptotic genes (Slc7a11/Slc3a2/Gpx4) in tumor cells and promotes Ctss secretion, which activates macrophage TLR2-NF-κB and IL-6 production. This feedforward loop suppresses lipid peroxidation, enabling ferroptosis evasion and radioresistance. Targeting Ephb4 restores ferroptosis sensitivity. This study demonstrates that injured TG neurons upregulate and release NPY, which acts via Y2R on neighboring nociceptors while recruiting and activating TG macrophages. Macrophage-derived TNF-α increases neuronal excitability, establishing a self-amplifying neuron-macrophage feedback loop that sustains local neuroinflammation and mechanical allodynia. Y2R blockade (BIIE0246), Npy2r deletion, or macrophage depletion robustly reverses established pain. Clinically, CSF (but not plasma) NPY is elevated in TN patients and correlates with pain intensity. This study reveals that GPR161 is upregulated in PBMCs of ARDS patients and correlates negatively with oxygenation and positively with inflammation. Myeloid-specific GPR161 knockout attenuates lung injury and inflammation in LPS- and sepsis-induced ALI mice. Mechanistically, GPR161 directly binds and negatively regulates C5aR1, which interacts with PKM2 to promote glycolytic reprogramming and inflammatory responses in macrophages. CUSABIO recombinant human C5aR1 protein was used in SPR assays to confirm the direct GPR161-C5aR1 interaction.

Using AFM, this study quantifies that human ovarian matrix stiffness increases with age and in pathological conditions including chemotherapy-induced POI, PCOS, and endometriosis, with stiff matrices impairing follicle development and hormone secretion. Mechanistically, TGFβ1 upregulates IL-11 in fibroblasts during ovarian aging, promoting collagen secretion via ERK1/2 signaling and matrix stiffening. Il11ra1 deletion reduces activated fibroblasts, matrix stiffness, and improves follicle reserve and fertility. siRNA nanoparticles targeting IL-11 reduce ovarian stiffness and restore fertility in aged rodents. Enterotoxigenic Bacteroides fragilis is enriched in gallbladder cancer tissues and promotes tumor progression via dual mechanisms: surface protein MltD binds host TMPRSS13 to activate JAK2-STAT3 signaling, while toxin BFT activates NF-κB-CXCL1 axis to recruit MDSCs and promote angiogenesis. Deletion of BFT or disruption of MltD-TMPRSS13 abolishes these effects. CUSABIO BFT antibody was used for Western blot detection of the toxin.

This study developed a shape memory microneedle (SMN) that switches from sustained to burst release upon mild heating-induced X-to-O shape recovery after a single transdermal insertion. FSH and hCG are dual-loaded in GelMA layers within X-shaped slots: outer FSH is released over 72 h via gel degradation, and after heating, O-shape recovery fractures inner GelMA to burst-release hCG within 24 h. In rabbits, a single overdose SMN achieves ovulation comparable to multiple syringe injections while eliminating hormone peak stimulation, significantly reducing vascular leakage, ascites, hemoconcentration, and multi-organ inflammation.
