Intra-assay Precision (Precision within an assay): CV%<8%
Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision (Precision between assays): CV%<10%
Three samples of known concentration were tested in twenty assays to assess.
To assess the linearity of the assay, samples were spiked with high concentrations of human IL-12/P40 in various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay.

This ELISA kit provides quantitative measurement of human IL-12B (IL-12 p40 subunit) in biological samples, supporting research into Th1-driven immune responses and inflammatory signaling. Because IL-12 p40 is produced by activated antigen-presenting cells and circulates both as free monomer and as part of IL-12 (p70) and IL-23 heterodimers, accurate quantification across diverse sample contexts requires carefully matched detection parameters.
Detection Range: The 7.8–500 pg/mL range captures IL-12 p40 levels spanning baseline serum concentrations in healthy individuals through the elevated levels observed in active infection, autoimmunity, and sepsis, minimizing the need for extensive sample dilution in most inflammatory disease studies.
Sensitivity: At 3.9 pg/mL, the kit detects IL-12 p40 in low-abundance settings such as unstimulated dendritic cell supernatants, early innate immune activation, or plasma from patients prior to clinical flare, where p40 levels may fall below 10 pg/mL.
Sample Compatibility: Validation across serum, plasma, and tissue homogenates enables researchers to correlate systemic IL-12 p40 levels with local tissue expression — critical for studying mucosal immunity, tumor microenvironments, or granulomatous inflammation within a single experimental framework.
Assay Efficiency: Requiring only 50–100 µL per well and 1–5 hours to complete, the kit accommodates limited clinical specimens and time-sensitive experiments involving primary monocyte or macrophage stimulation assays.
This kit is directly applicable to studies of inflammatory bowel disease, psoriasis pathogenesis, and anti-IL-12/23 therapeutic monitoring.
