IL-36α (IL-1F6) signals through the IL-1Rrp2/IL-1RAcP receptor complex to drive NF-κB and MAPK activation, making it a key mediator in epithelial and immune cell inflammatory responses. This tag-free recombinant mouse IL-36α encompasses the full-length sequence (1–160 aa) and demonstrates confirmed binding to recombinant murine IL-1Rrp2 across a 0.15–5 µg/mL range when immobilized at 1 µg/mL in functional ELISA, supporting its use as a coating antigen for antibody validation, a reference standard in cytokine detection assays, and a reliable ligand in receptor-binding studies. Endotoxin levels below 1.0 EU/µg minimize LPS-driven artifacts in sensitive cell-based assays such as NF-κB reporter activation, keratinocyte or immune cell stimulation, and in vivo murine inflammation models. Purity exceeding 95% by SDS-PAGE, combined with this stringent endotoxin control, provides a suitable basis for downstream signaling pathway dissection and antibody development workflows.
