B7-H6 functions as a stress-induced ligand on tumor cells and infected cells, engaging the activating receptor NKp30 on natural killer cells to trigger cytotoxicity—a pathway central to NK cell-mediated immune surveillance. This recombinant protein, spanning residues 25–262 and expressed in baculovirus, demonstrates specific antibody binding in functional ELISA with an EC50 of 10.90–12.22 ng/mL, confirming that the extracellular domain retains conformational epitopes required for receptor-ligand interaction assays and therapeutic antibody epitope mapping.
