Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Description

This Mouse Claudin-18 (Cldn18) recombinant protein was produced in mammalian cell, where the gene sequence encoding mouse Cldn18 (1-264aa) was expressed with the N-terminal 6xHis tag. The activity was validated. In addition, this mouse Cldn18 recombinant protein was developed through the Virus-Like Particles (VLPs) Platform. It is a four-pass transmembrane protein.

CLDN18 belongs to one of the tight junction protein family, and its isoform CLDN18.2 is highly expressed in a variety of tumor tissues, such as gastric cancer (60%-80%), pancreatic cancer (50%), esophageal cancer (30%-50%), and lung cancer (40%~60%), etc., but almost no expression in normal tissues, so it has excellent therapeutic potential. Even in the metastatic lesions of gastric cancer, CLDN18.2 has a high expression level, which makes CLDN18.2 a potential target for targeted therapy and immunotherapy.
CLDN18 was initially identified as a target gene of the transcription factor t/ebp/nkx2.1, consistent with its homology to other Claudin family members, and CLDN18 was later shown to localize at cellular tight junctions in mice and humans. The full-length murine CLDN18 protein is 264 amino acids, and its homology with human CLDN18.2 protein is as high as 90%. Its structure is similar to that of human. Both of them have four transmembrane domains: the intracellular NH2 terminus & COOH-terminus and two extracellular loops ECL1 & ECL2.
