The expression vector recombined with the recombinant DNA was transfected into the yeast cells for expression. The recombinant DNA resulted from the fusion of the gene coding for the 25-265aa of the Bovine APOA1 protein and the N-terminal 6xHis tag gene. The product was purified and isolated to get the recombinant Bovine APOA1 protein with N-terminal 6xHis tag. The purity of this recombinant Bovine APOA1 protein reaches up to 90%. Under SDS-PAGE condition, this recombinant APOA1 protein showed a band with a molecular weight of about 30 kDa on the gel.
APOA1 is a gene encoding a protein named Apolipoprotein A1 (abbreviated APOA1) and belongs Apolipoprotein A1/A4/E family. APOA1 is the major component of high-density lipoprotein (HDL) and plays a central role in reverse cholesterol transport. Because HDL is able to transport cholesterol and certain fats called phospholipids through the bloodstream from the body's tissues to the liver. A study has showed that apoB and apoA1 were simple clinical indicators, and the apoB/apoA1 ratio was closely related with CHD in the overweight group. The apoB/apoA1 ratio increase indicated an increased risk of CHD and may provide some useful information in the differential diagnosis.
