The synthesis of Recombinant H.pylori ureA protein began with the isolation of ureA gene from the native genome. The next step was to amplify the ureA gene by inserting the ureA gene into an expression vector and then introducing the vector into the selected host cell, E.coli. Significant amounts of recombinant H.pylori ureA protein were produced by the host only when expression genes were added. For the purpose of protein purification, the vector contains N-terminal 6xHis tag to the ureA DNA sequence. The purity of this protein is 85%+ determined by SDS-PAGE.
ureA (also known as hpuA) is a gene encoding a protein named urease subunit alpha (ureA) in Helicobacter pylori. The protein encoded by this gene is also known as urea amidohydrolase subunit alpha and belongs to urease gamma subunit family and urease beta subunit family. Its orthologous protein is known as the gamma/beta subunit (UreAB) in most other bacteria. This protein plays an important role in urea catabolic process.
