I'm wondering what the maximum concentration and lowest glycerol percentages you’ve achieved previously if you have at all?
They want to know if increasing the concentration and decreasing the glycerol in the storage buffer will influence the protein activity and stability?
For CSB-EP001853HU, previously, we sent lyophilized powder for most of the time, and the concentration before freeze-drying was 1.35mg/ml.
If deliver with liquid form, we can increase the concentration by 0.46mg/ml, and then send it after reconstitution.
Glycerol has a protective effect on proteins. Firstly, it can increase the stability of the protein. Secondly, it can lower the freezing point of the system and facilitate the preservation of protein at low temperatures. Especially when the protein is repeatedly frozen and thawed, protein damage (precipitated or inactivated) can be avoided as much as possible. As for the final concentration of glycerol, in principle, the final concentration ranges from 5% to 50% is OK, which may be related to the storage time. The protein may be unstable without glycerol.
When the protein is stable, the activity may not be affected. Regarding the activity of CSB-EP001853HU, I'm sorry that we haven't tested it for the time being and haven't received feedback from customers, so we can't guarantee 100% activity.
Can you increase the concentration of the human recombinant AOAH? The one we previously bought has a concentration of 0.46 mg/mL;
Can you lower the percentage of glycerol in the storage buffer? Currently, the buffer contains 50% glycerol.
