The expression of recombinant human WNT5A protein includes the construction of the expression vector containing the recombinant DNA and the transformation of the expression vector into the E.coli, which provides a variety of macromolecules and components required for transcription and translation. The recombinant DNA was formed by fusing the N-terminal 6xHis tag sequence to the designated sequence encoding the 62-380aa of the human WNT5A protein. This N-terminal GST-tagged recombinant human WNT5A protein is also characterized by high purity, >85%. Under SDS-PAGE condition, this recombinant WNT5A protein migrated to the band of about 40 kDa molecular weight. It may have applications in WNT5A-mediated cancer research.
WNT5B is considered to be a non-classical Wnt protein in the process of fat differentiation. Wnt protein family includes at least 19 members, which is a group of secretory glycoproteins rich in cysteine and participates in many biological processes such as embryonic development, cell proliferation and differentiation, and even tumor occurrence. Wnt5b is a member of the same family of proteins as Wnt5a. Evidence has accumulated that Wnt5b expression is involved in mouse chondrocyte maturation. Wnt5b were reported to be highly expressed in head and neck squamous cell carcinomas. Other studies also found highly expressed in breast cancer with high invasive activity. Of late, the role of Wnt5b overexpression in cancer aggressiveness has been identified. For example, in lung cancer and pancreatic cancer cells, upregulated Wnt5b has been observed, but the biochemical properties and cancer-associated activities of Wnt5b have not been well characterized.
