TRAIL (TNFSF10) selectively triggers apoptosis in transformed cells by engaging death receptors TNFRSF10A and TNFRSF10B, making it a critical tool for dissecting extrinsic apoptotic signaling in cancer models. This recombinant protein, spanning residues 114–281 of the mature extracellular domain, demonstrates receptor-binding competence with an EC50 of 3.897–5.215 ng/mL against immobilized TNFRSF10B in functional ELISA, confirming that the folded trimer retains native interaction geometry. The low endotoxin burden (< 1.0 EU/μg) supports use in cell-based apoptosis assays with primary tumor cells or cancer cell lines, where LPS contamination would confound interpretation of death-receptor-mediated versus TLR4-driven responses. Purity exceeding 90% by SDS-PAGE, combined with the validated receptor engagement, provides a suitable basis for signaling pathway studies examining caspase-8 activation, NF-κB modulation, and DISC assembly, as well as for antibody development workflows requiring a functional positive control.
