The DNA fragment encoding the 1-389aa of the humancoronavirus 229E Nucleoprotein (N) protein was fused with N-terminal 6xHis tag gene and then was inserted into the expression vector, which was subsequently transformed into the E.coli for expression. The resulting product was further purified to obtain the recombinant human coronavirus 229E Nucleoprotein (N) protein. The purity of this recombinant Nucleoprotein (N) protein is greater than 90% assessed by Bandscan software analysis combined with SDS-PAGE. This Nucleoprotein (N) protein showed a band on the gel with a molecular weight of approximately 55 kDa.
Studies indicated that N protein could interact with SARS and COVID-19. Among them, a study pointed that in patients with COVID-19, N antibodies have been observed, suggesting that N protein is capable of producing an immune response. To some extent, researchers revealed that N protein is very likely to preform additional roles during the viral infection. Besides, N proteins are served as inhibitors of interferon (IFN) in pathogenicity because N proteins were documented to antagonize the IFN response. Some researchers also discovered that N proteins of SARS-CoV-2 plays an important role in the inhibition of IFN-βproduction.
