We are using the E7 HPV protein from Lot# 03541 and need to change to a buffer without glycerol in order to make our experiments work. Is this possible? Or is the protein not H2O soluble?

A:
Thanks for your inquiry.
1. It is suggested that you can change the buffer by yourselves, by performing dialysis. This experiment is very simple and easy to operate.
You can perform dialysis according to the storage buffer in our Datasheet&COA, WITHOUT glycerol, ie 20 mM Tris-HCl, 0.5 M NaCl, pH 8.0, to dialyze with a solution of the appropriate volume.
However, there will be certain loss of protein after the dialysis process.
2. We haven't detected the stability of the protein in water. In general, proteins will be stable at a certain ion concentration and pH buffering conditions,
but we have also seen a protein that can be stably present in the water environment before (The stability of this protein also has not been detected yet).
3. If you do not have the appropriate conditions to deal with, you can also re-order the protein without glycerol. If you have interest, we will send you the details.

Target Background

Function
Plays a role in viral genome replication by driving entry of quiescent cells into the cell cycle. Stimulation of progression from G1 to S phase allows the virus to efficiently use the cellular DNA replicating machinery to achieve viral genome replication. E7 protein has both transforming and trans-activating activities. Induces the disassembly of the E2F1 transcription factor from RB1, with subsequent transcriptional activation of E2F1-regulated S-phase genes. Interferes with host histone deacetylation mediated by HDAC1 and HDAC2, leading to transcription activation. Plays also a role in the inhibition of both antiviral and antiproliferative functions of host interferon alpha. Interaction with host TMEM173/STING impairs the ability of TMEM173/STING to sense cytosolic DNA and promote the production of type I interferon (IFN-alpha and IFN-beta).
