The E7 protein possesses transformation activity and transactivation ability. Its structure comprises two functional domains: one is the N-terminal region containing the conserved Cys-X-X-Cys (CXXC) motif, responsible for interacting with specific proteins within the cell; the other is the C-terminal region, including the conserved LXCXE sequence, which binds to the Retinoblastoma protein (Rb) within the cell. By binding to the Rb protein, the E7 protein inhibits the function of Rb, leading to uncontrolled cell cycle and abnormal proliferation. This causes cells to lose their normal growth suppression mechanisms, promoting cellular transformation and carcinogenesis.
