ROR1 serves as a promising target in oncology and developmental neuroscience, yet its pseudokinase status and complex extracellular architecture demand native post-translational modifications for faithful ligand recognition. This mammalian-expressed fragment spanning residues 30–403 captures the extracellular domain and satisfies the purity and endotoxin criteria typical for therapeutic antibody screening, with greater than 95% purity by SDS-PAGE and less than 1.0 EU/μg endotoxin. Functional ELISA validation demonstrates specific antibody binding with an EC50 of 0.8551–0.9363 ng/mL when the protein is immobilized at 2 μg/mL, providing quantitative evidence that supports its use in competitive inhibition assays, blocking antibody screening, and epitope mapping studies. The confirmed binding activity makes this protein appropriate for SPR or BLI affinity characterization, small-molecule inhibitor screening, and as a positive control in receptor-ligand interaction platforms targeting ROR1-dependent signaling pathways.
