CUSABIO used the Rat Thyrotropin subunit beta (Tshb) DNA sequence encoding amino acids 21-132 to generate a fusion protein with an N-terminal 10xHis-tag and a C-terminal Myc-tag. The resulting recombinant full-length of mature protein was produced in the Baculovirus. It was subjected to SDS-PAGE (15% gel) under reducing conditions and presented a molecular mass band of about 19 kDa on the gel. The purity of the Tshb protein is greater than 85%. In addition to specific antibody synthesis, this recombinant Tshb protein may be used in the studies of Tshb-involved cancers.
Tshb, together with Tsha, makes up the thyrotropin (TSH). Tshb determines the specificity of TSH, which is responsible for the activation of the thyroid gland, playing a key role in controlling development and metabolism in mammals and other vertebrates. TSH is also involved in the specific developmental processes such as larval metamorphosis in amphibians, as well as larval and secondary metamorphoses in some teleost species. Additionally, TSH may regulate the functions in immune or reproductive systems through pleiotropic effects and multiple target tissues of thyroid hormones.
