  • IHC image of CSB-RA093733A0HU diluted at 1:100 and staining in paraffin-embedded human colorectal cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.
  • Overlay Peak curve showing Hela cells stained with CSB-RA093733A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.

Description

Cyclin B2 plays a central role in cell cycle progression, functioning as a regulatory subunit of cyclin-dependent kinases during the G2/M transition. As a key component of the mitotic machinery, CCNB2 accumulates during S and G2 phases before being rapidly degraded at the metaphase-anaphase transition, making it an important marker for studying cell proliferation, mitotic entry, and checkpoint regulation in both normal and malignant cells.

This recombinant monoclonal antibody offers the reproducibility and consistency that demanding research applications require. Generated through recombinant expression technology and derived from a single clone, it provides sequence-defined specificity that eliminates the lot-to-lot variability often encountered with traditional hybridoma-derived antibodies. The rabbit IgG format, combined with affinity chromatography purification, ensures high purity and reliable performance across experiments.

Validation studies demonstrate this antibody's utility across multiple experimental platforms. Immunohistochemistry performed on paraffin-embedded human colorectal cancer tissue shows clear detection of CCNB2 at dilutions between 1:50 and 1:200, with successful staining achieved using standard citrate buffer antigen retrieval. Flow cytometry analysis using HeLa cells confirms intracellular detection capability, with distinct positive signal separation from isotype control, supporting its application in cell cycle studies using this widely-used human cervical carcinoma model.

The validated performance in both tissue sections and cell-based assays makes this antibody well-suited for researchers investigating mitotic regulation, cancer cell proliferation, and cell cycle dynamics. Its compatibility with fixed and permeabilized samples provides flexibility for characterizing CCNB2 expression patterns in human tumor specimens and cultured cell populations.

Gene References into Functions
  1. Data show that Islet-1 (ISL1) activated the expression of cyclin B1 (CCNB1), cyclin B2 (CCNB2) and c-myc (c-MYC) genes by binding to the conserved binding sites on their promoters or enhancers. PMID: 27183908
  2. Data show that the cyclin B2 was overexpressed in bladder cancer tissue. PMID: 26706119
  3. overexpression of CCNB2 protein is associated with clinical progression and poor prognosis in non-small cell lung cancer PMID: 26349989
  4. Cyclin B1 and cyclin B2 are interchangeable for ability to promote G2 and M transition in HeLa cells. PMID: 24324638
  5. there was no significance in the 5-year overall survival rates among the patients with squamous cell carcinoma between expressing lower and higher level of cyclin B2 mRNA PMID: 24375198
  6. Suggest that cytoplasmic CCNB2 may function as an oncogene and could serve as a potential biomarker of unfavorable prognosis over short-term follow-up in breast cancer. PMID: 23282137
  7. The relative expression level of circulating CCNB2 mRNA in cancer patients was significantly higher than that in normal controls and benign diseases group. Circulating CCNB2 mRNA level significantly correlated with cancer stage and metastasis status. PMID: 21161946
