DFFA, also known as DFF45 or ICAD (Inhibitor of Caspase-Activated DNase), plays a central role in the execution phase of apoptosis. This 37 kDa protein functions as both an inhibitor and chaperone for the nuclease CAD, which is responsible for the characteristic DNA fragmentation observed during programmed cell death. When caspase-3 cleaves DFFA during apoptosis, it releases active CAD to fragment chromosomal DNA, making DFFA a critical marker for studying apoptotic pathways, cancer biology, and cell death mechanisms.
This recombinant monoclonal antibody, clone 4B12, offers the consistency and reliability that demanding research applications require. Developed using recombinant technology with a defined sequence, it eliminates the lot-to-lot variability that can compromise longitudinal studies and reproducibility. The rabbit IgG format, raised against a synthetic peptide derived from human DFFA, provides high specificity for human samples.
Validation data demonstrates robust performance across multiple experimental platforms. In western blot applications, the antibody detects a clean 37 kDa band in HepG2 whole cell lysate at dilutions of 1:500–1:2000, with the observed molecular weight matching the predicted size precisely. Immunohistochemistry validation in paraffin-embedded human colorectal cancer and stomach tissue sections shows clear staining at 1:50–1:200 dilutions using standard citrate buffer antigen retrieval. Flow cytometry analysis in HeLa cells confirms intracellular detection capability, enabling quantitative single-cell studies of DFFA expression.
This antibody serves researchers investigating apoptosis regulation, tumor biology, and therapeutic responses where programmed cell death plays a determining role in experimental outcomes.
