This study reveals a novel role of adrenomedullin 2 (ADM2), produced by enteric neurons, in promoting the tissue-protective functions of group 2 innate lymphoid cells (ILC2s) and limiting intestinal inflammation. Genomic deletion of ADM2 receptor subunits in ILC2s significantly reduces tissue-protective ILC2 responses and amphiregulin (AREG) production, increasing susceptibility to intestinal damage and inflammation. Conversely, therapeutic delivery of recombinant ADM2 induces AREG+ protective ILC2s and limits intestinal inflammation. Moreover, expression of ADM2 receptor genes is altered in patients with inflammatory bowel diseases, correlating with reduced AREG expression in ILC2s.
