This human programmed death 1 (PD-1) ELISA kit employs the quantitative sandwich enzyme immunoassay technique to measure the levels of human PD-1 in different samples, including serum, plasma, tissue homogenates, or cell lysates. The enzyme-substrate chromogenic reaction is also used to amplify the signal and quantify the levels of the analyte through the intensity of the colored product. The color intensity positively correlates with the amount of PD-1 bound in the initial step.
PD-1, also called PDCD1, is a type I transmembrane protein preferentially expressed in immune cells such as T, B, and NK cells. PD-1 and its ligand PD-L1 are critical immune checkpoint proteins that mediate inhibition of T cell proliferation, activation, cytokine production, altered metabolism, and cytotoxic T lymphocytes (CTLs) killer functions and eventual death of activated T cells. Tumor cells hijack these inhibitory pathways to escape host immune surveillance through overexpression of PD-L1. Antibodies targeting PD-1 have shown efficacy in increasing the anti-tumor activity of T cells in PD-L1-expressing cancers.
