CUSABIO produced the PD-1 recombinant monoclonal antibody by following the following steps: 1. Separation of B cells. Obtain B cells from the spleen of the immunized animal. During the immunization process, the recombinant human PD-1 protein is used as the immunogen. 2. RNA isolation and cDNA synthesis. Extract RNA from the B cells and then reversely transcribe it into cDNA. 3. Construction of recombinant vector. With cDNA as the template, the gene encoding the PD-1 antibody will be extended with a degenerate primer. The PD-1 antibody gene is inserted into the vector. 4. Introduce the vector into the host system. Transfection of the recombinant vector into the host cells for antibody expression. Harvest the PD-1 recombinant monoclonal antibodies from the cell culture supernatant and then purify them using affinity chromatography. 5. Antibody validation. This PD-1 recombinant monoclonal antibody has been tested to react with human PD-1 protein in ELISA.
