Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

Claudin-9 plays a critical role in tight junction assembly and barrier function, and its aberrant expression has been implicated in tumor progression and metastasis across multiple cancer types. This mammalian-expressed protein spanning residues 2–217 retains native post-translational modifications that support proper folding of the extracellular loops essential for homotypic and heterotypic claudin interactions. Functional validation demonstrates specific binding to an anti-CLDN6/9 recombinant antibody with an EC50 of 2.812–3.524 ng/mL in ELISA format, confirming that the extracellular epitopes remain accessible and correctly presented—a prerequisite for antibody development and validation studies targeting claudin family members. With purity exceeding 95% by SDS-PAGE and endotoxin levels below 1.0 EU/μg, this protein meets the quality thresholds commonly required for cell-based assays investigating tight junction dynamics, tumor cell migration and invasion, and the molecular mechanisms underlying claudin-mediated changes in epithelial barrier integrity.

Target Background

Function
Plays a major role in tight junction-specific obliteration of the intercellular space, through calcium-independent cell-adhesion activity.; (Microbial infection) Acts as a receptor for hepatitis C virus (HCV) entry into hepatic cells.
Gene References into Functions

  2. Claudins 6, 7, and 9 expressions are closely related to gastric carcinogenesis. PMID: 19960275
  3. Mouse studies suggest that claudin-9 deficiency may cause hearing loss not only in mice but also in humans. PMID: 19696885
  4. Residues N38 and V45 in the first extracellular loop (EL1) of CLDN9 are necessary for HCV entry. PMID: 17804490
  5. CLDN9, clustered with CLDN6 at human chromosome 16p13.3, is a four-transmembrane protein with WWCC motif, defined by W-X(17-22)-W-X(2)-C-X(8-10)-C. PMID: 12736707
