Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

This Recombinant Human Claudin-9 (CLDN9) is a premium, high-purity membrane protein fragment designed for functional and structural studies. Its key advantage lies in the specialized detergent-based purification process, which is essential for solubilizing and stabilizing this multi-pass transmembrane protein in its native, active conformation. Expressed in mammalian cells, the recombinant CLDN9 protein corresponds to amino acids 2-217 of human CLDN9 and is engineered with both an N-terminal 10xHis tag and a C-terminal Twin-Strep tag for highly efficient two-step purification.

The exceptional quality of this preparation is rigorously validated. SDS-PAGE analysis under reducing conditions confirms a purity greater than 95%, showing a clean, single band. Most importantly, its functional integrity is demonstrated through a quantitative binding ELISA: when immobilized, the protein exhibits specific, high-affinity interaction with an anti-CLDN6/9 recombinant antibody, yielding an EC50 of 2.812-3.524 ng/mL. This confirms that the extracellular domains are correctly folded and accessible. With low endotoxin levels (<1.0 EU/µg), this ready-to-use reagent provides an indispensable tool for demanding applications such as ligand-binding assays, antibody characterization, and structural studies targeting the native state of the CLDN9 protein.

Target Background

Function
Plays a major role in tight junction-specific obliteration of the intercellular space, through calcium-independent cell-adhesion activity.; (Microbial infection) Acts as a receptor for hepatitis C virus (HCV) entry into hepatic cells.
