Reconstitution

Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Description

Neuropilin-1 serves as a critical co-receptor for VEGF165 and semaphorins, making it a key target in angiogenesis research and cardiovascular biology. This recombinant human NRP1 encompasses the full-length mature extracellular domain of isoform 2 (aa 22–644) with a C-terminal hFc1 tag, and its mammalian cell expression ensures native-like glycosylation and proper folding of the CUB, coagulation factor, and MAM domains essential for ligand engagement. Functional ELISA confirms robust binding to immobilized human VEGF165 with an EC50 of 22.68–34.55 ng/ml, providing validated support for receptor-ligand interaction assays, competitive inhibition studies, blocking antibody screening, and affinity characterization by SPR or BLI. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the quality criteria typical for therapeutic antibody epitope mapping and small-molecule inhibitor screening in drug discovery workflows.

Mediates the chemorepulsant activity of semaphorins. Recognizes a C-end rule (CendR) motif R/KXXR/K on its ligands which causes cellular internalization and vascular leakage. It binds to semaphorin 3A, the PLGF-2 isoform of PGF, the VEGF165 isoform of VEGFA and VEGFB. Coexpression with KDR results in increased VEGF165 binding to KDR as well as increased chemotaxis. Regulates VEGF-induced angiogenesis. Binding to VEGFA initiates a signaling pathway needed for motor neuron axon guidance and cell body migration, including for the caudal migration of facial motor neurons from rhombomere 4 to rhombomere 6 during embryonic development. Regulates mitochondrial iron transport via interaction with ABCB8/MITOSUR.; Binds VEGF-165 and may inhibit its binding to cells. May induce apoptosis by sequestering VEGF-165.
