Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Shelf Life

The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.

Description

NKp30 (NCR3) is a key activating receptor on natural killer cells that recognizes stress-induced ligands on tumor and infected cells, making it a critical node in innate immune surveillance. This biotinylated cynomolgus macaque NCR3 extracellular domain (aa 19–135) demonstrates robust ligand-binding activity with an EC50 of 1.171–1.377 ng/mL against immobilized NCR3LG1 in functional ELISA, providing quantitative evidence of native conformational integrity suitable for ligand-receptor interaction studies and competitive inhibition assays. Mammalian cell expression preserves the post-translational modifications essential for authentic receptor folding, supporting applications in therapeutic antibody epitope mapping, blocking antibody screening, and affinity characterization by surface plasmon resonance or biolayer interferometry. The protein meets quality thresholds commonly required for immunology research, with purity exceeding 95% by SDS-PAGE and endotoxin levels below 1.0 EU/μg, while the C-terminal biotinylation enables oriented immobilization in binding assays and high-throughput screening platforms.

Function

Cell membrane receptor of natural killer/NK cells that is activated by binding of extracellular ligands including BAG6 and NCR3LG1. Stimulates NK cells cytotoxicity toward neighboring cells producing these ligands. It controls, for instance, NK cells cytotoxicity against tumor cells. Engagement of NCR3 by BAG6 also promotes myeloid dendritic cells (DC) maturation, both through killing DCs that did not acquire a mature phenotype, and inducing the release by NK cells of TNFA and IFNG that promote DC maturation.
