Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Shelf Life

The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Description

Tau protein plays a central role in microtubule stabilization and is a defining pathological hallmark of Alzheimer's disease and related tauopathies, making it an essential target in neuroscience research. This recombinant rat Mapt, covering the full-length mature sequence (aa 2–752) with an N-terminal 10×His tag positioned away from the microtubule-binding repeats, is expressed in mammalian cells to support native-like folding and post-translational modifications critical for cytoskeletal dynamics and polymerization studies. Functional ELISA confirms binding activity against an anti-MAPT recombinant antibody with an EC50 of 136.2–238.9 ng/mL, providing a validated basis for protein-protein interaction assays, antibody development, and use as a positive control in Western blot or immunofluorescence experiments. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the quality criteria typical for structural studies such as cryo-EM and crystallography.

Function

Promotes microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity. The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both. Axonal polarity is predetermined by tau localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. The short isoforms allow plasticity of the cytoskeleton whereas the longer isoforms may preferentially play a role in its stabilization.
