Within the E. coli expression system, the recombinant human tyrosine-protein phosphatase non-receptor type 11 (PTPN11) is synthesized through the incorporation of the gene fragment encoding the full-length mature PTPN11 protein (2-593aa) into a suitable expression vector. This vector is strategically designed to include an N-terminal 6xHis-tag downstream of the gene fragment, facilitating the convenient purification and detection of the recombinant protein. After introducing the recombinant plasmid into E. coli host cells and performing selection using appropriate markers, the transformed cells are cultured in a growth medium optimized for optimal protein expression. To initiate recombinant protein production, IPTG is added as an inducer, thereby inducing the expression of recombinant PTPN11 protein. Following a suitable incubation period, the cells are harvested and subjected to lysis, liberating the intracellular contents containing the expressed recombinant PTPN11 protein. The subsequent purification process is employed to isolate the recombinant protein, which exhibits a remarkable purity level of up to 90% as evaluated through SDS-PAGE analysis.
