Buffer

If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose.

Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Shelf Life

The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Description

Amino acids 1-1003 constitute the expression domain of recombinant Human MOV10. The theoretical molecular weight of the MOV10 protein is 119.6 kDa. This MOV10 protein is produced using e.coli expression system. The MOV10 coding gene included the N-terminal 6xHis tag, which simplifies the detection and purification processes of the recombinant MOV10 protein in following stages of expression and purification.

The human helicase MOV10 is a member of the UPF1-like RNA helicase family. MOV10 is involved in various cellular processes, including RNA metabolism and innate immunity. It plays a crucial role in RNA interference (RNAi) and RNA silencing pathways by unwinding RNA duplexes and facilitating the processing of small RNAs. MOV10 is also implicated in mRNA translation regulation, RNA editing, and retroviral restriction. Additionally, it has been associated with RNA-induced silencing complex (RISC) assembly and the control of endogenous retroelement expression. The multifaceted functions of MOV10 highlight its significance in maintaining RNA homeostasis and host defense mechanisms.

5' to 3' RNA helicase contributing to UPF1 mRNA target degradation by translocation along 3' UTRs. Required for microRNA (miRNA)-mediated gene silencing by the RNA-induced silencing complex (RISC). Required for both miRNA-mediated translational repression and miRNA-mediated cleavage of complementary mRNAs by RISC. In cooperation with FMR1, regulates miRNA-mediated translational repression by AGO2. Restricts retrotransposition of long interspersed element-1 (LINE-1) in cooperation with TUT4 and TUT7 counteracting the RNA chaperonne activity of L1RE1. Facilitates LINE-1 uridylation by TUT4 and TUT7. Required for embryonic viability and for normal central nervous system development and function. Plays two critical roles in early brain development: suppresses retroelements in the nucleus by directly inhibiting cDNA synthesis, while regulates cytoskeletal mRNAs to influence neurite outgrowth in the cytosol. May function as a messenger ribonucleoprotein (mRNP) clearance factor.
