Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

The gene encoding the Rhesus macaque CD40LG protein (44-261aa) is incorporated into a plasmid vector to generate recombinant plasmid, which is then introduced into e.coli cells. The e.coli cells containing the recombinant plasmid are selected and cultured under conditions that stimulate the expression of the gene of interest. A N-terminal 6xHis tag is attached to the protein. After expression, affinity purification is employed to isolate and purify the recombinant Rhesus macaque CD40LG protein from the cell lysate. Denaturing SDS-PAGE is utilized to resolve the resulting recombinant Rhesus macaque CD40LG protein, revealing a purity level exceeding 90%.

 Q&A
Q:

My only questions would be if this is designed to bind receptor in an ELISA. Also for me It cannot be fused to an Fc domain (The His tag is okay)
Any advise?

A:
Very nice to receive your inquiry. We don't have stock for this protein currently.
What we did with this protein was a complete extracellular domain, 44-261aa, which is a protein that is fused with 6xHis, not a Fc fusion protein.
A ligand containing a soluble form of 113-261aa could theoretically bind to a receptor in ELISA, but we have not verified what you are doing. Generally complete extracellular domain is suitable for many experiments.

Target Background

Function
Cytokine that acts as a ligand to CD40/TNFRSF5. Costimulates T-cell proliferation and cytokine production. Its cross-linking on T-cells generates a costimulatory signal which enhances the production of IL4 and IL10 in conjunction with the TCR/CD3 ligation and CD28 costimulation. Induces the activation of NF-kappa-B. Induces the activation of kinases MAPK8 and PAK2 in T-cells. Mediates B-cell proliferation in the absence of co-stimulus as well as IgE production in the presence of IL4. Involved in immunoglobulin class switching.; Acts as a ligand for integrins, specifically ITGA5:ITGB1 and ITGAV:ITGB3; both integrins and the CD40 receptor are required for activation of CD40-CD40LG signaling, which have cell-type dependent effects, such as B-cell activation, NF-kappa-B signaling and anti-apoptotic signaling.
Gene References into Functions
