Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Description

CD40 ligand plays a central role in T cell–B cell crosstalk, driving B cell activation, immunoglobulin class switching, and germinal center formation, making it a key target in cancer immunotherapy and autoimmune disease models. This mammalian-expressed protein, spanning residues 112–260 and carrying an N-terminal mFc1 tag, demonstrates robust receptor engagement with an EC50 of 5.1–6.1 ng/mL in functional ELISA against immobilized mouse CD40, confirming its capacity to trigger downstream signaling events. The low picomolar binding potency supports its use in B cell proliferation and activation assays, NF-κB and MAPK pathway studies, and in vivo tumor immunology models where CD40–CD40L interactions modulate dendritic cell maturation and anti-tumor immunity. With endotoxin levels below 1.0 EU/μg and purity exceeding 95%, this preparation meets the quality thresholds commonly required for primary immune cell cultures and antibody development workflows where LPS contamination would confound cytokine-specific responses.

Its cross-linking on T-cells generates a costimulatory signal which enhances the production of IL4 and IL10 in conjunction with the TCR/CD3 ligation and CD28 costimulation. Induces the activation of NF-kappa-B. Induces the activation of kinases MAPK8 and PAK2 in T-cells. Mediates B-cell proliferation in the absence of co-stimulus as well as IgE production in the presence of IL4.
