  • IHC image of CSB-RA439668A0HU diluted at 1:100 and staining in paraffin-embedded human glioma cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.

  • Overlay Peak curve showing Hela cells stained with CSB-RA439668A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.

Description

PPP2R5D serves as a critical regulatory subunit of protein phosphatase 2A (PP2A), one of the major serine/threonine phosphatases governing cell cycle progression, signal transduction, and DNA damage responses. This B56 delta isoform directs PP2A substrate specificity and subcellular localization, making it essential for understanding phosphatase-mediated regulation in both normal physiology and disease states. Mutations in PPP2R5D have been linked to neurodevelopmental disorders, positioning this target at the intersection of basic cell biology and translational research.

This recombinant monoclonal antibody, clone 18C3, offers the reproducibility that demanding experimental workflows require. Generated in rabbit and raised against a synthetic peptide derived from human PPP2R5D, the recombinant format ensures sequence-defined consistency across production lots, eliminating the variability that can compromise longitudinal studies or multi-site collaborations. Affinity chromatography purification delivers a clean reagent suitable for sensitive detection methods.

Validation data demonstrates reliable performance across multiple platforms. Immunohistochemistry staining in paraffin-embedded human glioma cancer tissue and small intestine tissue confirms utility for both oncology research and normal tissue characterization, with recommended dilutions of 1:50–1:200. Flow cytometry analysis using HeLa cells shows clear signal separation from isotype controls, enabling quantitative assessment of PPP2R5D expression at the single-cell level. The antibody is also validated for ELISA applications.

Whether investigating PP2A complex assembly, exploring phosphatase dysfunction in cancer, or characterizing PPP2R5D-associated developmental disorders, this antibody provides a dependable tool for researchers working with human samples across immunohistochemical and cytometric applications.
